用1例日本滑膜肉瘤(SS)患者的瘤组织标本接种裸鼠,获得了肿瘤生长。亲本肿瘤与裸鼠肿瘤在病理组织形态上存在一定差异,但两者的免疫组织化学特征相同。染色体分析表明,SS细胞株存在染色体数目和结构异常,患者外周血淋巴细胞染色体核型为正常女性,46,XX。 SS细胞株巴氏小体检出率低于对照,其正常X染色体比易位X染色体晚复制17小时。DNA印迹实验表明,SS DNA存在D2S3座位等位片段丢失,D1S57,D17S5和D13S30座位基因的部分缺失,但无DXS7,DXS14,和D2S44座位基因改变。  相似文献

Using a combined method of pulse-labelling trees and analysing detailed distribution of 13C tracer within tree rings, we studied how photo-assimilates incorporated on a given day are then distributed in a tree ring. A branch of a 4-year-old Cryptomeria japonica D.Don tree growing in Tsukuba, Japan was pulse-labelled with non-radioactive 13CO2 on two occasions: 29 May 2001 and 18 September 2001. Two discs were cut from the stem on 4 March 2002, one immediately under and the other 0.5 m below the branch and put through high-resolution δ 13C analysis. δ 13C peaks were observed in both the earlywood and latewood of the concerned tree ring, corresponding to each pulse-labelling date. The earlywood peaks was broader than the latewood peaks, possibly reflecting seasonal variation of the width of wood developing zone. Half-widths of the peaks were measured and used as indicators for the potential time resolution of tree-ring isotope analysis. The half-widths of the peaks indicated a time resolution no finer than 8.7–28 and 33–42 d in the early and latewood, respectively. Holocellulose extraction yielded only a slight change to the shape of the δ 13C peaks. 13C tracer pulse-labelled in May and September reached tangentially different locations in the lower disc, suggesting a seasonal change in the pathway of carbohydrates. Local consumption of spring assimilates and long-distance downward transport of autumn assimilates were also suggested.  相似文献

Several cell types migrated cut from small pieces of newt testes cultivated in vitro. Flat fibroblastic cells migrated out within a few days. Then, secondary spermatogonia, identified by the presence of germ cell-specific substances and by the shape and appearance of their nucleus and subcellular organelles, migrated out over the sheet of fibroblastic cells. Sertoli cells co-migrated with secondary spermatogonia, maintaining a similar cellular arrangement to that of testicular cells in vivo. Mitosis of secondary spermatogonia both in clusters and as single cells was frequent from the third day until about 2 weeks after inoculation. During mitosis, active and periodic rotation of chromosomes was observed. Identification of the cell types and studies on their behavior were performed by electron microscopy and phase contrast microscopy.  相似文献

We attempted to determine natural agonists of GPR39 in fetal bovine serum (FBS). FBS was conditioned to extract peptides and fractionated by two types of HPLC. The activity of each fraction was monitored by intracellular calcium mobilization. Then the purified active ingredient was analyzed by inductively coupled plasma mass spectrometry. In this fashion, Zn2 + ion was identified as an agonist of GPR39, though no peptidergic molecules were found. The calcium-mobilizing activity of Zn2 + was not abolished by pertussis toxin but was by a phospholipase C (PLC) inhibitor, U73122, indicating that the activity of GPR39 is mediated through the Gqα -PLC pathway. In addition, Zn2 + also activated mouse and rat GPR39, showing that the function of GPR39 as a Zn2 + receptor is conserved across species. This study is the first exploration of GPR39 agonists in FBS and indicates that GPR39 functions as a Gq-coupled Zn2 +-sensing receptor.  相似文献
